В промоторной области гена FAS находится однонуклеотидный (single nucleotide polymorphism) полиморфизм -670A/G, связанный с заменой нуклеотида аргинина на гуанин, который ассоциирован с предрасположенностью к системной красной волчанке, рассеянному склерозу, саркоидозу и аутоиммунному гепатиту.</p><p>Цель – проверить в российской выборке больных гипотезу о возможной ассоциативной связи полиморфизма -670А/G гена FAS с предрасположенностью к системной склеродермии (ССД), ее клиническим и аутоиммунным фенотипами.</p><sec><title>Материал и методы</title><p>Материал и методы. В исследование включено 90 пациентов с ССД, которые были классифицированы по клиническим и аутоиммунным фенотипам. Контрольная группа состояла из 152 условно здоровых неродственных лиц, сопоставимых по полу и возрасту. Полиморфизм -670 A/G гена FAS был изучен с помощью полимеразной цепной реакции с последующим анализом полиморфизма длин рестрикционных фрагментов.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. На сравнительно небольшой выборке российских больных не установлено статистически значимой ассоциативной взаимосвязи изученного полиморфизма -670A/G с предрасположенностью к ССД в целом и большинством ее клинико- иммунологических фенотипов. Выявлена статистически значимая положительная ассоциативная связь аллели G (генотипы -670GG и GA) с наличием дигитальных язвочек и хроническим течением заболевания. У больных с интерстициальным поражением легких (ИПЛ) аллель G (генотипы -670GG и GA) выявлялась реже, чем у больных без ИПЛ.</p></sec><sec><title>Заключение</title><p>Заключение. Наши данные показывают, что полиморфизм FAS -670A&gt;G играет роль в предрасположенности к некоторым клиническим фенотипам ССД у российских больных.</p></sec></abstract><trans-abstract xml:lang="en"><p>The FAS antigen (Apo-1/CD95) is a key molecule of apoptosis in most cell types, including activated immune cells and fibroblasts. The FAS gene promoter region contains a single-nucleotide polymorphism (-670A/G) associated with a substitution of the nucleotide arginine for guanine, which is associated with the predisposition to systemic lupus erythematosus, multiple sclerosis, sarcoidosis, and autoimmune hepatitis.</p><sec><title>Objective</title><p>Objective: to test the hypothesis that the FAS -670А/G polymorphism may predispose to systemic sclerosis (SS), its clinical and autoimmune phenotypes in a Russian patient sample.</p></sec><sec><title>Subjects and methods</title><p>Subjects and methods. The instigation enrolled 90 SS patients who were classified according to clinical and autoimmune phenotypes. A control group consisted of 152 apparently healthy unrelated individuals matched for sex and age. The FAS -670А/G polymorphism was studied by polymerase chain reaction, followed by restriction fragment length polymorphism analysis.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. A relatively small sample of Russian patients showed no statistically significant association of the studied FAS -670 A/G polymorphism with the predisposition to SS as a whole and the majority of its clinical and immunological phenotypes. There was a statistically significant positive association of the G allele (the FAS -670 GG+GA genotype) with the presence of digital ulcers and the chronic course of the disease. The G allele (the FAS -670 GG+ GA) was detected less frequently in patients with interstitial lung disease (ILD) than in those without ILD.</p></sec><sec><title>Conclusion</title><p>Conclusion.
